In this grant application, we aim to molecularly decipher how the receptor for advanced glycation endproducts (RAGE) binds to nucleic acid ligands and interacts with itself. We will define the crystal structure of the RAGE/DNA complex and test the function of critical residues involved in binding of nucleic acids. We will further investigate how ligand binding to RAGE leads to cellular activation and influences its trafficking behavior. In a mouse respiratory syncytial virus infection model we will finally assess the physiological role of RAGE in lung antiviral responses.
