Systemic impact of TGR5 in liver damage and regeneration (B01)

Bile acids (BAs) and their receptors, such as FXR, TGR5 and S1PR2 are key regulators during liver damage and liver regeneration. In the past, project B01 characterized the function and regulation of the BA receptor TGR5 in non-parenchymal cells of the liver, in macrophages and in intestinal epithelial cells. We demonstrated that TGR5-deficient mice are more susceptible towards cholestatic and inflammatory liver damage as compared to their wildtype littermates. The projects aims to characterize the role of TGR5 in liver, intestinal and immune cells during liver damage and regeneration and to investigate the crosstalk of TGR5 with other BARs, especially S1PR2. Both TGR5 and S1PR2 are druggable targets with a variety of potential applications in liver diseases including hepatobiliary cancers. A better understanding of the regulation, function and interaction of TGR5 and S1PR2 during liver damage/ regeneration will provide important information for novel therapeutic approaches.
