Combining advanced fluorescence methods and nanoagents (B03)

In this project, we use single molecule methods to observe and improve the functionality of individual nanoagents while also using nanoagents to provide complementary capabilities or to enhance different fluorescence methods. For the next funding period, we will continue to investigate the DNA roboarm (Simmel A02) and utilize metal enhanced fluorescence to increase the temporal resolution with FRET (Tinnefeld A13). Using light, we will control the trafficking of motors and generate artificial lipid rafts (Lohmüller A08/ Seshold B09). In the third part, we will use multimodal, coherent Raman imaging (Ploetz, Junior Group leader) as chemically sensitive analysis tool for porous nanoparticles and combine it with 3D Orbital Tracking, to track nanoagents in living systems without fluorescent labeling or photobleaching.
