Final Report Abstract

FACS sorting was used to obtain discrete B cell subpopulations ranging in maturity from naive B cells to memory B cells in a study of DNA methylation programming during B cell maturation. These subpopulations were then used to observe a broad epigenetic programming of selective transcription factor binding sites coincident with the degree of B cell maturation. Comparisons were made between normal B cells and malignant B cells from 268 patients with chronic lymphocytic leukemia (CLL). Postoperative ileus (POI), the most frequent complication after intestinal surgery, depends on dendritic cells (DCs) and macrophages. FACS and cell sorting was used for their quantification and selection in a study that showed that CD103+CD11b+ DCs and the intestinal microbiome are a prerequisite for the activation of intestinal monocytes and macrophages and for dysregulating intestinal motility in POI. FACS was used to quantify the number of liver resident macrophages in a study to investigate the role of bile acids and their receptor farnesoid X on macrophage proliferation and response to viral infection in hepatocytes.
