This project demonstrated that overall gene expression changes after closed head TBI strongly depend on post-traumatic hematoma formation. Our data also show that IKK/NF-B signaling in glia cells is a critical player in the orchestration of the post-traumatic neuroimmune response and how astrocytic NF-B may interfere with blood-brain barrier homeostasis. On the basis of these findings we aim to further characterize the role of NF-B in the bi-directional brain-immune communication after TBI. Because prolonged IKK/NF-B signaling in oligodendrocytes disturbed myelin maintenance, we also intend to identify NF-B regulated effectors triggering TBI-induced white matter loss.
