Vesicular transport in eukaryotic cells underlies a modular organization with defined submodular activities that are functionally conserved for each transport process, irrespective of donor and target membrane. In previous work, we have initiated a structural and biochemical characterization of the multi-subunit tethering complex TRAPP (transport protein particle) and its regulation by protein palmitoylation. It is proposed to extend these studies towards a comprehensive description of the three-dimensional structure, assembly and function of TRAPP which may serve as a paradigm for tethering complexes in vesicle transport in general.
