The endothelial plasma membrane is dynamically remodeled through demand-driven exocytosis of endothelial-specific storage compartments, the Weibel-Palade bodies (WPB). We study how cargo is delivered from endolysosomes to WPB, how cell stress alters this poorly characterized pathway, and analyze the impact on PM hubs. We focus on endolysosomal cholesterol balance, and thus also use and further develop novel cholesterol analogs (CHIMs) as chemical tools to investigate cholesterol-related dynamic membrane processes.
