In the previous funding period we found that a breakdown of the intestinal vascular barrier plays a crucial role in the pathogenesis of IBD. Here we will analyze the temporal relation of vessel barrier breakdown and disease development, investigate the molecular mechanisms of inflammation-induced endothelial barrier disruption and determine the angiocrine impact of the inflamed intestinal vasculature on epithelial barrier functions. Finally, we will assess the therapeutic potential of pharmacological blood vessel stabilization in IBD. The ultimate goal of this project is to provide new perspectives for vessel-directed therapy approaches for IBD.
