Identifizierung und Charakterisierung von Influenza Virus aktivierenden Proteasen in der menschlichen Lunge
Zusammenfassung der Projektergebnisse
Vorarbeiten zeigten, dass Influenza-A-Viren (IAV) des Menschen durch die zelluläre Protease TMPRSS2 aktiviert werden. Mehrere TMPRSS2-verwandte Proteasen wurden ebenfalls als IAV-Aktivatoren identifiziert, aber ihre Expression in relevanten Zielzellen und ihr Beitrag zur viralen Ausbreitung waren unklar. Außerdem waren die Struktur der TMPRSS2-mRNA und geeignete Angriffspunkte für Antisense-Oligonukleotide (ASOs) unbekannt. Darüber hinaus war unklar, inwieweit und über welchen Mechanismus die „Halsdomäne“ von TMPRSS2 zur Virusaktivierung beiträgt. Schließlich war ungeklärt, ob TMPRSS2 die virale Ausbreitung in Primaten fördert. Diese Fragen sollten durch das vorgeschlagene Forschungsvorhaben geklärt werden. Im Hinblick auf die COVID-19-Pandemie wurde die Forschungsrichtung beibehalten, aber die Arbeiten konzentrierten sich auf SARS-CoV-2, da bekannt war, dass auch Coronaviren durch TMPRSS2 aktiviert werden. Tatsächlich konnten wir nachweisen, dass SARS-CoV-2 TMPRSS2 für den Zelleintritt nutzen kann. Außerdem setzten wir erfolgreich Chemikalien ein, um Einblicke in die Struktur der TMPRSS2-mRNA zu gewinnen, was das erfolgreiche Design von ASOs ermöglichte. Darüber hinaus fanden wir heraus, dass mehrere TMPRSS2-verwandte Proteasen zusammen mit dem SARS-CoV-2-Rezeptor ACE2 in den Atemwegen koexprimiert werden und somit die virale Ausbreitung unterstützen könnten. Zusätzlich erhielten wir Hinweise darauf, dass die Halsregion für die Aktivierung von IAV und Coronaviren durch TMPRSS2 und verwandte Proteasen wichtig ist, der zugrunde liegende Mechanismus konnte jedoch nicht geklärt werden. Schließlich identifizierte wir TMPRSS2- Inhibitoren und lieferten Hinweise darauf, dass TMPRSS2 (oder verwandte Proteasen), aber nicht die Hilfsprotease Cathepsin L, für die Ausbreitung von SARS-CoV-2 im menschlichen respiratorischen Epithel ex vivo erforderlich ist.
