Molecular function of uracil phosphoribosyltransferase (UPP) in chloroplast proteostasis and its effect on acclimation (B08)

UPP is active in the salvage pathway and exhibits a moonlighting activity in chloroplast biogenesis. The project focuses on the identification of the molecular function of UPP in chloroplast proteostasis by testing already identified interaction partners (like PetC) by complementation approaches and direct assays (BIFC, CoIP). The entire UPP interactome will be identified through the combination of TurboID, Complexome analysis, forward genetics and the pamiR approach, in both Arabidopsis and Chlamydomonas, complemented by proteome studies of de-etiolation and light series experiments. Moreover, co-crystallization of UPP with interacting proteins is planned.
