In this grant proposal, we apply for an mass spectrometer to be used in the biological institutes (Institute for Applied Biosciences, Botanical Institute) at the Karlsruhe Institute of Technology (KIT). Applications comprise both analyses of metabolites (including (non-protein-)polymers) as well as proteomics applications. A hybrid quadrupole mass spectrometer, as a high-resolution, accurate-mass, sensitive and sufficiently fast scanning spectrometer, is the ideal device fulfilling the requirements as defined by the research areas of the participating working groups. The mass spectrometer is to be used, among other things, in shotgun proteomics of complex samples (food, cells and tissues, biological fluids), in the analysis of post-translational protein modifications, in structure elucidation of glycoconjugates (directly or after enzymatic hydrolysis) and plant secondary metabolites as well as for highly sensitive and accurate quantification approaches using targeted techniques (including stable isotope dilution assays). In order to create ideal conditions for analyses of metabolites as well as for proteomics applications, the mass spectrometer needs to be coupled to either an ultra-high performance liquid chromatography system (UHPLC) or a nano high performance liquid chromatograph (NanoH-PLC). The system is completed by ion sources including electrospray ionization (ESI) and atmospheric chemical ionization (APCI) probes and a nitrogen generator with compressor.
