Hepatic DIO1 affects systemic and local TH metabolism. Impaired TH signalling negatively affects micro- and macronutrient metabolism, contributing to hyperlipidaemia, fatty liver, fibrosis and eventually carcinogenesis. DIO1 transcription and translation are controlled by endo- and exogenous modifiers, including sex, age, cytokines, selenium, and drugs. We hypothesise that hepatic DIO1 expression can be controlled and increased via a better understanding of and active interference with these modulating factors, thereby enhancing local hepatic TH action and preserving liver health.
