Role of local TH action in epilepsy (P19)

Previously we have unraveled an aberrant balance of glutamatergic versus GABAergic neuro-transmitter systems in Mct8/Oatp1c1 DKO mice and highlighted a pronounced seizure susceptibility of these mice similar to AHDS patients with MCT8 deficiency. We will now advance on the long-term consequences of seizures in the Mct8/Oatp1c1DKO brain as well as dissect developmental from adult requirements of local TH signaling for seizure susceptibility using inducible TR knockout models. Likewise, we will assess if developmental versus adult-onset TH analogue treatment improves seizure susceptibility. Finally, we will answer, if common anti-seizure drugs affect the brain thyroidal state thereby causing central hypothyroidism in patients.
