This project will dissect how oncogenic FLT3-ITD and cKIT-D816V drive tissue factor (TF) expression to promote immune escape in AML. We will first elucidate the oncogenic signalling pathways downstream of FLT3-ITD and cKIT-D816V that drive TF expression in AML cells. We will then analyze the mechanisms by which TF mediates immune escape of leukaemia cells in vitro and in vivo. Finally, we will validate our findings from mouse models in human AML using GWAS, mass cytometry, microclot assays and kinase inhibition approaches.
