In this project, we will use a chemically programmable CAR (cpCAR) that can be redirected to target multiple antigens simultaneously or sequentially to address heterogeneous target cell populations and to prevent antigen loss as a major obstacle to effective CAR-T cell therapy. We will perform careful analysis of target cell engagement, cpCAR signaling and functional output in comparison to ‘conventional’ scFv-based CARs in the context of hematologic and solid tumors. Further, we will use the cpCAR as a co-receptor to augment co-stimulatory signaling and in logic gates to enhance selectivity for target cells.
