T cell engineering to overcome immune dysregulation induced by acute leukemia (B04)

Development of combinatorial engineering strategies to enhance efficacy of T cell immunotherapy while reducing off-target effects and overcoming inhibitory cellular circuits. This will be achieved by utilizing virus-free CRISPR/Cas9-guided CAR knock-in and site-specific genomic integration of transcription factors. By implementing promoter-specific control of transgene expression, we anticipate improved longevity of T cell immunotherapy, ultimately aiming for improved therapeutic outcomes.
