Subproject B06 develops an innovative strategy for cell-based targeted drug delivery to combat infections at otherwise inaccessible implant sites. Macrophages, pre-loaded in vitro with detection and therapeutic systems, migrate to sites of infection, where controlled cargo release is triggered by external stimuli such as ultrasound or alternating magnetic fields. Building on systems developed during the first funding period, now multiplexing of antibacterial agents, including bacteriophages, will be explored. To enable autonomous, stimulus-responsive drug release, infection-induced signaling cascades will be integrated into the carrier macrophages. These approaches will be complemented by pharmacokinetic and pharmacodynamic analyses.
