Project C01 will investigate the crosstalk between canonical FcgRs and the C-type lectin receptor Mincle in the context of inflammatory bone loss. Depending on ligand availability, independent sets of receptors may compete for the common FcRg-chain to trigger downstream activation of tyrosine-protein kinase SYK and phosphoinositide (PI)3 kinase to generate osteoclasts. The long-term goal of the project is to create an interaction map of FcRg-chain dependent cell surface receptors expressed during inflammatory osteoclastogenesis allowing identifying positive and negative checkpoints involved in inflammatory bone remodelling.
