The potential use of human stem cells in regenerative and reparative medicine is highly promising, but holds many challenges. One of the most important of these is the establishment of culture conditions for stem cells that reproduce in vitro selfrenewal and reprogramming events which are observed only rarely in vivo. There are now indications that the rich media used as the basis of most in vitro culture systems to date may not be well-suited to the metabolism of stem cells. Based on the results of preliminary work in the applicant's laboratory, a systematic optimisation of basic culture conditions for the propagation of stem cells will be undertaken, together with analyses of the relationship between stem cell metabolism, stress responses, and differentiation potential. The aim of this work is to define growth conditions appropriate to the high efficiency in vitro amplification and directed differentiation of primary stem cells.
