The project investigates the effect of CD19-CAR-T cell therapy on the molecular skin phenotype in systemic sclerosis, a severe autoimmune disease with high mortality characterised by skin and organ fibrosis. The aim is to deeply phenotype the effects of CD19-CAR-T cell therapy on fibrotic tissue remodelling and vasculopathy using Spatial omics (MERSCOPE) and imaging mass cytometry. Based on these findings, the project will identify mediators that regulate B cell-induced changes in the skin and will identify molecular treatment response patterns as basis to improve personalized treatment strategies in the future.
