This project focuses on the mechanisms how astrocyte-derived lipoproteins, particularly APOE, contribute to amyloid deposition in cerebral amyloid angiopathy (CAA). It explores whether APOE aggregates act as nucleation sites for β-amyloid (Aβ) within the walls of blood vessels. Using transgenic mice, spatial localization techniques, and lipidomic profiling, the project aims to clarify the site and mechanism of APOE aggregation, the contribution of lipid components, and the therapeutic potential of targeting aggregated APOE to slow disease progression in CAA.
