Autoantibodies are major drivers of tissue inflammation. Importantly, however, autoantibodies may be present years before the initiation of clinical disease, suggesting that checkpoints must be in place directly regulating autoantibody activity. A B cell intrinsic factor that has the ability to modulate both, the pro- and anti-inflammatory activity of IgG, is antibody glycosylation, which will be the focus of project 13 in the third funding period. We will investigate how the B cell intrinsic antibody glycosylation network is regulated during B cell development and how checkpoint control proteins, infections and therapeutic interventions modulate the B cell glycosylation machinery.
