Functional impact of MALT1 protease activation in B cell lymphomagenesis (P07)

MALT1 acts as a scaffold and a protease in lymphomas addicted to oncogenic activation of BCR signalling. While MALT1 scaffolding activates NF-κB, the contribution of MALT1 protease activity to lymphomagenesis is still unclear. We demonstrated that genetic destruction of TRAF6-MALT1 interaction in mice leads to loss of MALT1 scaffolding, but increased MALT1 protease function. Here, we want to analyse how specific activation of MALT1 protease alone or in combination with NF-κB driver mutations (e.g. oncogenic MYD88) is triggering B cell activation and expansion during different stages of B cell development. We will unravel the molecular consequences of MALT1 protease activation and evaluate options for therapeutic inhibition.
