The mechano‐genomic code: pioneer factors as decision makers (P04)

This project wants to decipher the mechano-specific pioneer transcription factor (TF) code in endothelial cells. They will integrate several next-generation sequencing-based assays to understand genomic regulation of fluid shear stress (FSS)-induced cell shape changes and identify genomic loci bound by SMAD and pioneer TF complexes. They will then evaluate the mechano-responsitivity of those composite motifs in luciferase reporter gene assays. By comparing different cell types subjected to the same mechanical trigger, these mechano-reporters will ultimately allow for assessing the common mechano-genomic pioneer TF code.
