GTPases, proteins that bind and hydrolyze guanosine 5´-triphosphate (GTP), are key regulators of a variety of cellular processes. Examples are: transfer of extracellular information into the cell, ribosomal protein synthesis, translocation of polypeptides at the endoplasmic reticulum, intracellular vesicle transport, endocytosis, exocytosis, actin polymerization and other cytoskeletal functions, cell cycle control, nuclear transport, as well as cell growth and differentiation. This priority programme aims at combining the efforts of the national laboratories working on GTPases to accelerate the dynamic developments in this area of research. This cooperation allows to resolve questions that cannot be addressed by individual laboratories. The main goals of the programme are (i) to establish a general understanding of the structural and functional properties of GTPases as wells as of the molecular mechanisms of their interaction with associated regulatory proteins and (ii) to elucidate the functional principles of GTPase cascades designed to regulate complex cellular functions.
